The production of culture and capture fishery of Indian major carps and Exotic CARPS, biochemical analysis has been conducted in farmed reared composite species of Indian major carps and Exotic carps to study its biodiversity. General protein from gill, muscle, heart and kidney were separated in SDS-PAGE (10% gradient) in which no polymorphism was observed. The Rm values were critically analysed using Jaccard's coefficient and the dendogram generated on UPGMA basis by computer software gives the closeness among the species, but the overall observation found more or less to be tissue specific.

The purpose of this work was to assess reference values of serum biochemical indices (enzymes, metabolites and minerals) in some representative fish species belonging to Acipenseridae, Cyprinidae, Esocidae and Salmonidae family in order to establish a baseline data which will serve to monitoring nutritional-metabolic balance and healthy condition of these species from aquatic biocenosis or intensive exploitations. Serum samples were analyzed for glucose, total proteins, albumine, urea nitrogen, creatinine, uric acid, triglycerides, cholesterol, ALT, AST, GGT, alkaline phosphatase, amylase, total bilirubin, Ca, P, Mg, Fe, using a FullyVet automated chemical analyser. The obtained results were compared with those from other papers regarding serum biochemical profile of fresh water fish species. Eff ects of high water cadmium sulphate hydrate (3CdSO4 * 8H2O) (CdSO4) levels on the expression of serum proteins and tissue histopathology of the European chub (Leuciscus cephalus) was investigated. Three groups of fish (control, 1 st , and 2 nd), each containing 10 animals, were placed in separate tanks containing no, 1 mg/L, and 2 mg/L CdSO4, respectively, for 10 days. At the end of the study period, blood samples were taken and sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) was performed. Fish tissue samples were taken then routinely processed and stained with hematoxylin and eosin to observe histopathological changes. Depending on the increasing concentration of CdSO4, a gradual decrease in protein bands was determined. Inhibition of 35.3 kilodalton (kD) and 100.5 kD proteins in the 1st group and of 44.5 kD and 47.3 kD proteins in the 2 nd group was detected. Microscopic examination revealed degeneration in the secondary lamellar epithelia, hydropic degeneration and necrosis in chloride cells of the fish gill in CdSO4 given groups. Hydropic degeneration and necrosis were also observed in light to severe levels in the liver depending on the groups. It was concluded that CdSO4 has toxic eff ects on the expression of serum proteins and causes degenerative changes on gill and liver morphology in Leuciscus cephalus.

Elde edilen elektroforegramda, Orthrias angorae bureschi ve Orthrias panthera’da 13 sarkoplazmik protein bandi elde edildi. Bunun yanisira, bu baliklarin 5 sarkoplazmik protein bandinin ayni molekuler agirlikta oldugu bulundu ve benzerlik katsayisi (SC): 0.384 olarak bulundu. 117 goosefish plasma from the Mediterranean coast have been analysed. Total plasma proteins gave mean values of 2410.9 for males and 3. 3~t l. l gjl00 for females. The cellulose acetate and starch-gel electrophoresis showed 7 and 13 fractions respectively. Different albumin patterns were observed but their distribution did not have a genetic basis. In 107 specimens a mean TIBC value of 278*125pg Fe% was found. The saturation coefficient of the transferrin was from 15-20%. The specimens had a single Tf band and one individual had a slower Tf band. The presence of an haptoglobin polymorphism has been suggested. The plasma haptoglobin concentration by an electrophoretic method was determined and a concentration from 28-31 mg Hp% was obtained.

Dissolved organic nitrogen (DON) represents the least understood part of the nitrogen cycle. Due to recent methodological developments, proteins now represent a potentially characterisable fraction of DON at the macromolecular level. We have applied polyacrylamide gel electrophoresis to characterise proteins in samples from a range of aquatic environments in the Everglades National Park, Florida, USA. Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) showed that each sample has a complex and characteristic protein distribution. Some proteins appeared to be common to more than one site, and these might derive from dominant higher plant vegetation. Two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) provided better resolution; however, strong background hindered interpretation. Our results suggest that the two techniques can be used in parallel as a tool for protein characterisation: SDS-PAGE to provide a sample-specific fingerprint and 2D-PAGE to focus on the characterisation of individual protein molecules. Information on the structure and character of immunoglobulin of fishes is essential in health management. A study was carried out to characterize the serum immunoglobulin (IgM) of the Indian major carp, rohu Labeo rohita (Ham.). Rohu (500 g) were immunised with bovine serum albumin (BSA) and the anti-BSA antibody was purified employing BSAeCL agarose affinity column chromatography. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of the purified Ig in a 3% gel under non-reduced conditions revealed a single protein having a molecular weight of 850 kDa. Analysis of the purified serum in 10% SDS-PAGE under reduced conditions revealed that the immunoglobulin contained heavy and light chains with molecular weights of 85 and 23 kDa, respectively. A polyclonal mouse anti-rohu IgM was prepared and used in an immunodot test which showed a specific reaction of the crude rohu anti-BSA antiserum and the purified anti-BSA IgM with BSA. Results indicate that the immunoglobulin of L. rohita is tetrameric IgM, similar to that of other fishes. ª

An experiment was conducted to determine the utilization by mirror carp of yeast (PY), bacterial (MB), algal (SA), soybean (SY), casein (CS), and herring (HM) meal proteins as the sole protein sources in semi-purified rations. A low casein (CS LOW) diet was included in the trial so that true digestibility, Net Protein Utilization (NPU) and Biological Values (BV) could be determined. All the feeds contained 30% protein other than the fishmeal and bacterial diets which contained 25 and 35%, respectively. Specific growth rates (%/day) found were MB 2.78 > CS 2.4, HM 2.26 > PY 2.08 > SY 1.24, SA 1.18 and CS LOW 1.13. Food conversion was optimal with diet MB (1.14) followed by CS (1.39), HM (1.42), PY (1.55) < SA (2.50) < SY (2.86) and CS LOW (3.04). Protein efficiency ratio was highest with the low casein diet at 3.24 decreasing in the order HM 2.82, CS 2.48 > PY 2.08 > SY 1.35 and SA 1.15. All the test proteins were well digested, assimilation values ranging from 80.3% (HM diet) to 96.6% (PY diet). The NPU's and BV's calculated from body analysis data were respectively for each protein, HM 64 and 79%, MB 49 and 52%, CS 49 and 52%, PY 41 and 49%, SY 42 and 51%, SA 36 and 41%. The above biological values are compared with various chemical scores based on amino acid analysis of the test proteins. All the results are discussed fully and also compared to values previously obtained with identical diets in rainbow trout experiments.
Some biochemical characteristics of common carp (Cyprinus carpio) and silver carp (Hypophtalmichthys moltrix) surimi prepared by a conventional washing method and protein isolated using alkaline-acid-aided processes were investigated. Solubility of protein in silver carp and common carp were found to be highest by using the conventional washing method. Decreases in myoglobin and lipid contents in both fish were found in the alkaline-or acidaided process when compared to the conventional process (p<0.05). The percentage of protein recovery with the alkaline-aided process of silver carp was highest. The whiteness score in the alkaline solubilization process was higher than acid solubilization process and conventional method in silver carp. This result was different than the result obtained in common carp. There was no significant difference between treatments regarding total sulfhydryl (SH) groups (p>0.05). In protein patterns of Sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE), the lowest intensity of the myosin heavy chains (MHC) band was found in silver carp by the conventional washing process.

The objective of this study is to identify fi sh protein markers for detecting multiple species based on a comparative proteomic approach that relies on fragments with identical sequences. The possibilities and challenges of the use of peptides obtained from carp (Cyprinus carpio) and herring (Clupea harengus) proteins are discussed. A bioinformatic analysis was followed by an LC-MS/MS experiment to identify markers predicting the presence of fi sh allergenic proteins. Selected myosin peptides were found in carp protein hydrolysates with known sequences and in herring protein hydrolysates with unknown sequences. The results obtained for carp and herring proteins myosin and parvalbumin indicate that proteins with unknown sequences can be identifi ed by peptide markers. Such markers can be designed by disregarding the principle that peptides should be unique (present in one sequence). The challenge is to determine a group of proteins that can be detected by peptide identifi cation.
